【Overview】

A luciferase reporter cell line is a genetically engineered cell system designed to study gene expression and cellular signaling pathways in real-time. These cell lines are stably transfected with a reporter construct where the gene of interest's regulatory elements (e.g., promoter, enhancer, or response elements) control the expression of the luciferase enzyme. 【How It Works】

The cell line is exposed to experimental conditions (e.g., drugs, cytokines, or genetic manipulations).

If the pathway of interest is activated, transcription factors bind to the regulatory elements, driving luciferase expression.

Luciferase catalyzes the oxidation of luciferin in the presence of ATP and oxygen, emitting detectable light.

The light signal is measured using a luminometer or imaging system, with intensity proportional to transcriptional activity.
